Effect of Clostridium difficile toxin A on human intestinal epithelial cells: induction of interleukin 8 production and apoptosis after cell detachment.

چکیده
Clostridium difficile is the aetiological agent of pseudomembranous colitis, and animal studies suggest the essential role of secreted toxin A in inducing disease. This study examined the biological responses to toxin A by human intestinal epithelial cells. Confluent monolayers of Caco2, HT29, and T84 cells and primary epithelial cells in organ cultures of human colonic biopsy specimens and after detachment with EDTA were studied. Interleukin 8 was assayed using enzyme linked immunosorbent assay (ELISA). Purified C difficile toxin A induced cell rounding and detachment of monolayers of the epithelial cell lines. Cells in detached monolayers initially remained viable while adherent to each other. Subsequently, an increasing number of apoptotic cells appeared in suspension. Exposure to toxin A for 24 hours induced interleukin 8 production in T84 and HT29 cells. Toxin A also induced epithelial cell rounding, detachment, and apoptosis in organ cultures of human colonic biopsy specimens. During culture (in medium only), EDTA detached colonic epithelial cells produced interleukin 8 and cell death occurred by apoptosis. Colonic disease by C difficile may be initiated by toxin A mediated induction of epithelial cell interleukin 8 production and apoptosis after cell detachment from the basement membrane. Studies on isolated (toxin untreated) colonic epithelial cells suggest that interleukin 8 production and apoptosis occur as a consequence of cell injury and detachment.

clostridium difficile has been recognized as the major cause of pseudomembranous colitis.this bacterium produces two toxins(an enterotoxin -cytotoxin and a potent cytotoxin called toxin A and toxin B erespectively).these toxins have implicated in pathogenesis of the disease.however, histopathological effects of their molecular mass less than 100KDa have been essayed.in the persent study,we exami...

It is known that the major etiologic agent of pseudomembranous colitis in man is Clostridium difficile. With respect to traditional use of almond paste in the treatment of infantile diarrhea, we studied the effects of the aqueous extract of Amygdalus communis (AEAC) on the growth and toxin production of Clostridium difficile in culture medium and the rabbit ligated ileal loop. Three groups...
متن کاملP-156: Survey of In Vitro Effect of Resveratrol on Apoptosis of Human Endometrial Epithelial Cells
Background: In apoptosis is physiological cell death which eliminates injured and old cells. Any disruption in apoptosis, leads to disease. Resveratrol is a natural polyphenol with estrogen-like, antioxidant and antiinflammatory properties; also it showed cell prolifratory and inhibitory effect. The aim of present study was to determine resveratrol effect on apoptosis of human endometrial epith...

Clostridium difficile is an opportunistic pathogen inhabiting the human gut, often being the aetiological agent of infections after a microbiota dysbiosis following, for example, an antibiotic treatment. C. difficile infections (CDI) constitute a growing health problem with increasing rates of morbidity and mortality at groups of risk, such as elderly and hospitalized patients, but also in popu...
متن کاملMAPK-activated protein kinase 2 contributes to Clostridium difficile-associated inflammation.
Clostridium difficile infection (CDI) results in toxin-induced epithelial injury and marked intestinal inflammation. Fecal markers of intestinal inflammation correlate with CDI disease severity, but regulation of the inflammatory response is poorly understood. Previous studies demonstrated that C. difficile toxin TcdA activates p38 kinase in tissue culture cells and mouse ilium, resulting in in...
